It looks like it. There are conflicting reports out there as to whether Sheen has already signed or is still in negotiation. Director Marc Webb (500 Days of Summer) is still casting the movie and according to Deadline Hollywood, Sally Field may even show up as Aunt May.  So far, IMDB has the following cast list posted:

Andrew Garfield as Peter Parker / Spiderman

Emma Stone as Gwen Stacy (and not Mary Jane, surprisingly)

Rhys Ifans as Dr. Connors/The Lizard.  

 

We will get to see Rhys in action on November 19 when he shows up in Harry Potter Deathly Hallows Part 1 as Luna Lovegood’s daddy, Xenophilius.  About working on the Spiderman reboot, Rhys told the Associated Press that he is “very, very excited… It’s a great script, great director, and the money ain’t bad.”
